INCB052793 is a selective small molecule inhibitor of Janus kinase 1 (JAK1), developed for the treatment of advanced malignancies and hematologic cancers. By specifically binding to and inhibiting JAK1 phosphorylation, it disrupts JAK-dependent signaling pathways that are implicated in cellular proliferation and survival in tumor cells. Preclinical studies demonstrated anti-tumor activity both as a single agent and in combination with other anti-myeloma therapies. Clinical trials evaluated its efficacy in patients with advanced solid tumors, multiple myeloma, acute myeloid leukemia (AML), and myelodysplastic syndrome (MDS). However, development was discontinued after phase 2 due to lack of clinically meaningful responses[1][5][6][7][8].
