Overview

IND126 is an investigational small molecule inhibitor developed by Vegen Therapeutics that specifically targets the KRAS G12C mutation. KRAS G12C is a common oncogenic driver in several malignancies, most notably non-small cell lung cancer (NSCLC) and colorectal cancer (CRC), where a glycine-to-cysteine substitution at codon 12 results in a protein that is constitutively active in its GTP-bound state. IND126 is designed to covalently bind to the cysteine residue of the KRAS G12C mutant protein while it is in its inactive GDP-bound conformation. This binding locks the protein in the inactive state, thereby inhibiting downstream signaling pathways such as MAPK/ERK that drive tumor cell growth and survival. The drug is currently being evaluated in early-phase clinical trials for patients with advanced solid tumors harboring the G12C mutation.
