ITA-LNP is an experimental nanotherapy consisting of itaconate (ITA) conjugated to lipid nanoparticles (LNPs). Developed by researchers at Case Western Reserve University, it is designed to target atherosclerotic plaques and regulate lipid metabolism by stabilizing the cholesterol transporter ABCA1. In preclinical studies, ITA-LNP has been shown to reduce plaque size and increase ABCA1 protein levels in macrophages by preventing its degradation through the HO-1-calpain axis, thereby promoting cholesterol efflux. This dual-action approach addresses both inflammation and lipid accumulation within the arterial wall, representing a potential first-in-class treatment for atherosclerosis.
