Itacitinib + pembrolizumab is an investigational combination therapy consisting of itacitinib, a selective Janus kinase 1 (JAK1) inhibitor, and pembrolizumab, a monoclonal antibody targeting programmed death-1 (PD-1). Itacitinib modulates interferon signaling by inhibiting JAK1, potentially reducing immune suppression in the tumor microenvironment. Pembrolizumab blocks PD-1 on T cells to enhance anti-tumor immune responses.
