Overview

ITGβ4 siRNA is a small interfering RNA (siRNA) designed to silence the expression of integrin subunit beta 4 (ITGβ4), a junctional protein involved in endothelial barrier stability. In research settings, specifically in human pulmonary artery endothelial cells (HPAECs), ITGβ4 siRNA is utilized to investigate the molecular mechanisms of physiologic and pathologic shear stress. Studies indicate that knockdown of ITGβ4 leads to degraded endothelial barrier function and prevents cell alignment in the direction of flow, highlighting its role as a downstream effector of flow-sensitive transcription factors like KLF4. This reagent is primarily used as a laboratory tool to elucidate the pathophysiology of conditions such as high cardiac output pulmonary hypertension.
