J5dLPS + OMP is a novel vaccine construct designed to elicit broad-spectrum immunity against Gram-negative bacteria. It consists of detoxified, O-polysaccharide side chain-deficient lipopolysaccharide (J5 dLPS) derived from Escherichia coli 0111:B4 (J5 mutant), non-covalently complexed with the outer membrane protein (OMP) of Neisseria meningitidis group B. The removal of the immunodominant O side chain exposes highly conserved core glycolipid epitopes shared among Gram-negative bacteria. The Neisseria-derived OMP acts as a Toll-like receptor 2 ligand and stabilizes the glycolipid structure. Immunization with this construct induces high-titer polyclonal antibodies targeting these conserved core regions, promoting clearance of diverse Gram-negative pathogens and providing protection in animal models against lethal infections such as Francisella tularensis and Klebsiella pneumoniae. In veterinary applications, immunization in cows leads to high titers of anti-endotoxin antibodies that are transferred into colostrum, potentially reducing endotoxemia-related inflammation in recipients[1][2].
