JAB-23400 is an orally bioavailable, small molecule KRASmulti inhibitor developed by Jacobio Pharmaceuticals. It is designed to potently inhibit the activity of multiple KRAS mutants, including G12D, G12V, G12R, G12S, G12A, G13D, and Q61H, while maintaining selectivity over HRAS and NRAS. Notably, JAB-23400 targets KRAS in both its active (ON) and inactive (OFF) states, potentially offering broader therapeutic coverage than first-generation KRAS inhibitors that primarily target the OFF state. Preclinical data presented at the 2023 AACR Annual Meeting demonstrated its efficacy in various solid tumor models, particularly those of the digestive system. As of early 2024, the program is in the preclinical stage, with an Investigational New Drug (IND) application planned for the first half of 2024.
