JAB-23E73 is an investigational, orally administered small molecule inhibitor that targets multiple mutant forms of KRAS, a key oncogenic driver in many solid tumors. Unlike earlier KRAS inhibitors that are mutation-specific (such as those targeting only G12C), JAB-23E73 is described as a "pan-KRAS" inhibitor because it can inhibit both the active ("ON") and inactive ("OFF") states of various KRAS mutants—including G12D, G12V, G12A, G13D, Q61H—and shows good selectivity over HRAS and NRAS. Preclinical data indicate potent activity against KRAS-dependent cell lines and tumor regression in multiple xenograft models with favorable pharmacokinetic properties. The drug is being developed by Jacobio Pharmaceuticals for the treatment of advanced solid tumors harboring KRAS alterations[1][2][4][5][8].
