Overview

Jaktinib + azacitidine is an investigational combination therapy comprising the pan-Janus kinase (JAK) inhibitor jaktinib and the hypomethylating agent azacitidine. Jaktinib, developed by Suzhou Zelgen Biopharmaceuticals, is a small molecule that inhibits JAK1, JAK2, and JAK3, thereby modulating the JAK-STAT signaling pathway which is frequently dysregulated in myeloproliferative neoplasms. Azacitidine is a nucleoside analogue that inhibits DNA methyltransferases, leading to DNA hypomethylation and the reactivation of tumor suppressor genes. This combination is primarily being studied for the treatment of myelofibrosis, aiming to provide synergistic clinical benefits in patients who have had inadequate responses to JAK inhibitor monotherapy.
