Jing Guan Fang (JGF) is a Traditional Chinese Medicine (TCM) decoction primarily developed for the prevention of SARS-CoV-2 infection and the treatment of cold-like symptoms. Comprised of five medicinal herbs, including *Forsythia suspensa* and components like baicalin (from *Scutellaria baicalensis*), JGF acts through multiple pathways to inhibit viral entry and modulate the host immune response. Mechanistically, it induces the lysosomal-dependent degradation of the Angiotensin-Converting Enzyme 2 (ACE2) receptor and suppresses the expression of Transmembrane Protease Serine 2 (TMPRSS2), effectively blocking the fusion and entry of coronaviruses into host lung cells. Furthermore, JGF serves as an immunomodulator by regulating the JAK/STAT3 and MAPK/JNK signaling pathways; it exhibits a dual effect, promoting basal macrophage activity while suppressing lipopolysaccharide-induced inflammatory cytokine production, such as IL-6 and TNF-α. The formula also possesses antioxidant properties, leveraging flavonoids like baicalein to scavenge reactive oxygen species and mitigate inflammation.
