Overview

JNJ-4496 (formerly C-CAR039) is an investigational, dual-targeting autologous CAR T-cell therapy designed to bind both CD19 and CD20—two cell surface proteins commonly expressed on malignant B-cells. It incorporates a 4-1BB costimulatory domain intended to improve T-cell persistence and potentially address resistance mechanisms in relapsed or refractory large B-cell lymphoma. The therapy is under development for patients with relapsed or refractory large B-cell lymphoma, including diffuse large B-cell lymphoma, who have not previously received CAR T-cell therapy. Early clinical data (Phase 1b) show high overall and complete response rates with a promising safety profile[2][3][5].
