JNJ-95566692 is a novel, subcutaneous trispecific monoclonal antibody developed by Janssen Research & Development for the treatment of B-cell non-Hodgkin lymphoid malignancies. It functions as a T-cell engager by simultaneously binding to two distinct B-cell lineage antigens, CD79b and CD20, and the CD3 epsilon subunit on T cells. This dual-targeting approach for B-cell antigens is intended to enhance tumor specificity and mitigate the risk of immune escape through antigen loss, which is a common resistance mechanism in therapies targeting a single antigen like CD20. By cross-linking malignant B cells with cytotoxic T cells, JNJ-95566692 induces T-cell activation and targeted lysis of the lymphoma cells. It is currently being evaluated in Phase 1 first-in-human clinical trials both as a monotherapy and in combination with costimulatory agents like JNJ-87801493.
