KAD102 is a novel biologic drug developed to block the prolactin receptor, thereby preventing cancer cell growth signals and inducing autophagy—a process of cellular self-digestion that leads to cancer cell death. It primarily targets ovarian cancer by inhibiting the Jak2 signaling pathway crucial for cancer cell proliferation and downregulating GST enzyme expression associated with chemotherapy resistance. This mechanism potentially re-sensitizes chemo-resistant tumors to treatment. The drug is being developed as a longer half-life molecule compared to its predecessor (KAD101), aiming for improved activity and efficacy with less frequent dosing. Early clinical data from daily injectable formulations showed tumor reduction in ovarian cancer patients with a clean safety profile. The developer plans Phase 1 clinical trials starting in 2025.
