Overview

KITE-363 is an investigational autologous chimeric antigen receptor (CAR) T cell therapy engineered to target both CD19 and CD20, two antigens highly expressed on B-cell malignancies. The therapy uses a bicistronic vector to express two distinct CARs within the same T cell—one targeting CD19 with a CD28 costimulatory domain and another targeting CD20 with a 4-1BB costimulatory domain. This dual-targeting approach aims to overcome resistance mechanisms such as antigen escape, which can occur with single-antigen therapies, particularly in large B-cell lymphoma (LBCL). Preclinical studies have shown that KITE-363 can recognize and eliminate tumor cells expressing either or both antigens. It is being developed for relapsed or refractory B-cell lymphomas, including diffuse large B-cell lymphoma (DLBCL), mantle cell lymphoma, chronic lymphocytic leukemia, acute lymphocytic leukemia, Burkitt lymphoma, primary mediastinal B-cell lymphoma, and refractory acute myeloid leukemia[1][2][3][4][5].
