Drug intelligence / Profile preview
KRAS-G12D-mrRNA is a first-in-class, mutation-specific DNA methylation regulatory RNA (mrRNA) designed to selectively silence the oncogenic KRAS G12D mutant gene [PT1.03.03]. Developed by researchers at Zhongshan Hospital, Fudan University, as part of their 'OncoMutation Silence' epigenetic therapy platform, the drug utilizes a novel lncRNA-SNRPA/DNMT1-guided precise DNA methylation mechanism. It contains a motif that recognizes the KRAS G12D mutation and binds to the SNRPA/DNMT1 complex, thereby recruiting DNA methyltransferase 1 (DNMT1) to selectively upregulate DNA methylation at the mutant locus. This targeted epigenetic modification leads to the reversible suppression of KRAS G12D expression and inhibits tumor cell proliferation [PT1.03.03]. Formulated in lipid nanoparticles (LNPs) for intravenous administration, KRAS-G12D-mrRNA has demonstrated robust in vivo anti-tumor efficacy in preclinical models of KRAS-driven non-small cell lung cancer (NSCLC) [PT1.03.03].
