KRAS G12V T cell engager (A*03) is a bispecific T-cell engager (BiTE) being developed by Myrio Therapeutics (MyriO Tx). It is designed to target the KRAS G12V mutation, a common oncogenic driver in various solid tumors, specifically when the mutant peptide is presented by the Human Leukocyte Antigen (HLA) allele A*03 on the surface of tumor cells. The therapeutic molecule is engineered to simultaneously bind to the KRAS G12V/HLA-A*03 complex and the CD3 receptor on T cells, facilitating the formation of an immunological synapse. This interaction leads to the localized activation of T cells and the subsequent targeted lysis of cancer cells. The drug is currently in the discovery phase for the treatment of lung, pancreatic, and colon cancers.
