Overview

KT209 is a novel trispecific antibody (tsAb) that simultaneously targets CD19, CD20, and CD3, utilizing a 2:1:1 molecular format. Developed by Kintor Pharmaceutical, the agent is designed to redirect endogenous T cells to eliminate B-cell malignancies while maintaining a lower cytokine release profile compared to existing bispecific T-cell engagers (BiTEs) like blinatumomab or glofitamab. By co-engaging two distinct B-cell antigens (CD19 and CD20), KT209 aims to mitigate resistance caused by antigen escape or heterogeneous antigen expression, which are common limitations in single-antigen targeting therapies. Preclinical studies in leukemia cell lines and cynomolgus monkeys have demonstrated potent cytotoxicity and deep B-cell depletion, supporting its clinical development for relapsed or refractory B-cell diseases, including acute lymphoblastic leukemia.
