KT6-971 (also known as S-556971) is a small molecule cholesterol absorption inhibitor being developed by Kotobuki Pharmaceutical in collaboration with Shionogi for the treatment of dyslipidemia. It is described as a next-generation cholesterol absorption inhibitor that works by inhibiting the uptake of cholesterol in the small intestine. This mechanism typically involves the inhibition of the Niemann-Pick C1-like 1 (NPC1L1) protein, which is responsible for the transport of dietary and biliary cholesterol across the intestinal lumen. The drug has reached Phase 2 clinical development in Japan and Phase 1 in Europe.
