Overview

**L19-IL2 + rituximab** is a combination therapy consisting of the immunocytokine L19-IL2 and the monoclonal antibody rituximab. **L19-IL2** is a recombinant fusion protein composed of the human monoclonal antibody L19, which targets the extra domain B (EDB) of fibronectin (a marker of angiogenesis), fused to interleukin-2 (IL-2), an immune-stimulating cytokine. This fusion selectively delivers IL-2 to the tumor microenvironment, enhancing local immune activation. **Rituximab** is a monoclonal antibody targeting CD20 on B cells and mediates B cell depletion through immune mechanisms. When used together, these agents both target B-cell non-Hodgkin lymphoma by increasing immune cell infiltration and cytotoxicity within lymphoma tumors. Preclinical and early clinical evidence demonstrates that the combination has synergistic anti-lymphoma activity, inducing remission in xenograft models and clinical responses in relapsed/refractory B-cell lymphoma[1][2][4].
