Overview

Lanoconazole is a topical imidazole antifungal small molecule developed by Nihon Nohyaku in collaboration with Tsumura. It was approved in Japan in 1994 for the treatment of dermal mycoses, including tinea pedis (athlete's foot), tinea corporis, and tinea cruris. The drug exerts its therapeutic effect by inhibiting the enzyme lanosterol 14-alpha-demethylase (CYP51), which is a critical component of the ergosterol biosynthesis pathway in fungi. The depletion of ergosterol and the accumulation of methylated sterol precursors disrupt the fungal cell membrane structure and function. Lanoconazole exhibits potent activity against a broad range of pathogenic fungi, particularly Trichophyton and Candida species, and is typically administered as a 1% cream or solution.
