LCAR-AIO is a chimeric antigen receptor T cell (CAR-T) therapy developed by Legend Biotech. It consists of human T lymphocytes genetically modified with a lentiviral vector to express a tri-specific CAR targeting the tumor-associated antigens CD19, CD20, and CD22. This design enables the engineered T cells to recognize and eliminate B-cell malignancies that express any of these three antigens, potentially overcoming antigen escape mechanisms seen in prior single-target CAR-T therapies. Preclinical and early clinical data suggest that LCAR-AIO can eradicate antigen-heterogeneous B cell tumors, enhance expansion and persistence of CAR-T cells, and may be effective in patients who have relapsed after previous CAR-T treatments. The primary indications under investigation include relapsed/refractory B-cell lymphoma and other B-cell malignancies[1][3][6].
