LCAR-L19D (also known as JNJ-80801540) is an investigational dual-targeting chimeric antigen receptor T-cell (CAR-T) therapy designed to treat B-cell malignancies, including various forms of non-Hodgkin lymphoma (NHL) and chronic lymphocytic leukemia (CLL). The therapy is engineered to simultaneously target two distinct antigens, CD19 and CD20, which are widely expressed on the surface of malignant B-cells. This dual-targeting approach is intended to enhance therapeutic efficacy and mitigate the risk of disease relapse caused by antigen escape, a common resistance mechanism where cancer cells lose the expression of a single targeted antigen. Developed through a collaboration between Legend Biotech and Janssen Biotech (a Johnson & Johnson company), the process involves the ex vivo genetic modification of a patient's own T cells to express CARs specific for both targets. These modified cells are then expanded and re-infused into the patient to identify and eliminate cancerous B-cells. While the program reached Phase 1/2 development under the partnership, Janssen opted not to exercise its option to continue co-development in 2022, leaving Legend Biotech to proceed with the asset.
