LCB36 is a preclinical-stage bispecific antibody-drug conjugate (BsADC) being developed by LigaChem Biosciences (formerly LegoChem Biosciences) for the treatment of B-cell hematologic cancers, including B-cell lymphomas. LCB36 consists of a bispecific antibody simultaneously targeting B-lymphocyte antigen CD20 and B-lymphocyte antigen CD22, site-specifically conjugated via LigaChem's proprietary ConjuAll technology to a masked pyrrolobenzodiazepine (PBD) dimer prodrug payload. By targeting both CD20 and CD22, LCB36 is designed to overcome tumor antigen heterogeneity and reduce minimal residual disease (MRD), which are common drivers of relapse in B-cell malignancies. Upon binding and internalization, the PBD payload is selectively activated within cancer cells to induce DNA cross-linking and subsequent cell death.
