Overview

lcLNP anti-CD20 CAR mRNA is an experimental in vivo chimeric antigen receptor (CAR) therapy candidate developed by NanoVation Therapeutics. It utilizes the company's proprietary long-circulating lipid nanoparticle (lcLNP) platform to deliver mRNA encoding a CAR specific for the CD20 antigen. Unlike conventional LNPs that are rapidly cleared by the liver, lcLNPs are engineered for extended circulation and robust accumulation in extrahepatic lymphoid tissues, such as the spleen and bone marrow. Upon intravenous administration, the lcLNPs transfect immune effector cells (including T cells and myeloid cells) in situ, leading to the transient expression of the anti-CD20 CAR and subsequent depletion of CD20-positive B cells. This approach aims to provide the therapeutic benefits of CAR-T therapy without the complexities, high costs, and logistical challenges associated with ex vivo manufacturing.
