lcLNP-CAR mRNA is a preclinical-stage in vivo CAR T cell therapy platform developed by NanoVation Therapeutics. It utilizes proprietary long-circulating lipid nanoparticle (lcLNP™) technology to deliver mRNA encoding chimeric antigen receptors (CARs) directly to immune cells within the body, bypassing the need for complex ex vivo manufacturing. Unlike conventional LNPs that primarily target the liver, the lcLNP platform is engineered for extended circulation (up to 18 hours in NHPs) and functional delivery to extrahepatic tissues, including the bone marrow, spleen, and lymphoid organs, without the use of targeting ligands. Preclinical studies have demonstrated that intravenous administration of lcLNP-CAR mRNA can achieve near-complete B cell depletion at low doses (0.1 mg/kg) in humanized mice and non-human primates, suggesting potential applications in B cell malignancies and autoimmune disorders such as systemic lupus erythematosus (SLE).
