Overview

Lerociclib (G1T38) + osimertinib (Tagrisso) is a combination therapy being investigated for EGFR mutation-positive non-small cell lung cancer (NSCLC). Lerociclib is an oral CDK4/6 inhibitor designed to stop cancer cell growth by inhibiting the enzymes CDK4 and CDK6, which are involved in cell cycle progression. Osimertinib is an irreversible EGFR tyrosine kinase inhibitor that blocks EGFR-sensitizing and EGFR T790M-resistance mutations. The combination aims to enhance efficacy and potentially overcome resistance mechanisms in patients who have progressed on first-line EGFR inhibitors.
