Overview

Lm-LLO-CD105A (also known as ADXS31-171) is an investigational cancer vaccine consisting of a live-attenuated *Listeria monocytogenes* (Lm) bacterial vector engineered to secrete a fusion protein of truncated Listeriolysin O (tLLO) and the tumor-associated antigen CD105 (Endoglin). CD105 is a proliferation-associated antigen highly expressed on the endothelial cells of the tumor vasculature, making it a strategic target for anti-angiogenic immunotherapy. The Lm-LLO platform facilitates the delivery of the antigen directly into antigen-presenting cells, where it is processed and presented to stimulate a robust, polyfunctional CD8+ T-cell response. This immune response is designed to selectively destroy the tumor's blood supply, thereby inhibiting tumor growth and metastasis. While clinical development by Advaxis was terminated after Phase 1 trials in advanced solid tumors, the vaccine continues to be evaluated in preclinical research, particularly in combination with novel immunoadjuvants like ISG15.
