Overview

**Loncastuximab tesirine + mosunetuzumab** is a combination of two targeted immunotherapies developed for the treatment of relapsed or refractory diffuse large B-cell lymphoma (R/R DLBCL). Loncastuximab tesirine is an antibody-drug conjugate (ADC) targeting CD19 on B cells, delivering a cytotoxic pyrrolobenzodiazepine (PBD) payload (tesirine) that crosslinks DNA and induces cell death. Mosunetuzumab is a bispecific T-cell engager monoclonal antibody that simultaneously binds CD20 on B-cells and CD3 on T-cells, redirecting T-cells to kill CD20+ malignant B cells. The combination is under clinical investigation (e.g., phase 2 LOTIS-7, NCT05672251) to assess safety and efficacy in R/R DLBCL, leveraging dual targeting of tumor antigens and immune cell engagement to enhance tumor control[1][5][6].
