Overview

LS104 is a small molecule kinase inhibitor developed by LymphoSign Inc. that functions as a non-ATP-competitive inhibitor of Janus kinase 2 (JAK2) and Fms-like tyrosine kinase 3 (FLT3). Unlike traditional ATP-competitive inhibitors, LS104 binds to a site distinct from the ATP-binding pocket, which may offer improved selectivity and reduced off-target effects. It has been investigated primarily for the treatment of acute myeloid leukemia (AML) and JAK2V617F-positive myeloproliferative disorders (MPD). By inhibiting JAK2 autophosphorylation and downstream signaling, LS104 induces apoptosis in leukemic cells. It was notably the first non-ATP-competitive kinase inhibitor to enter clinical trials, specifically a Phase I study for patients with JAK2V617F-positive MPDs.
