LUCAR-G79D is an autologous chimeric antigen receptor (CAR) T-cell therapy being developed by Legend Biotech for the treatment of severe autoimmune diseases, including relapsed or refractory systemic lupus erythematosus (SLE) and idiopathic inflammatory myopathies (IIM). The therapy utilizes a dual-targeting construct that simultaneously recognizes B-lymphocyte antigen CD19 and B-cell maturation antigen (BCMA). By targeting these two antigens, LUCAR-G79D aims to achieve comprehensive depletion of both B cells and long-lived plasma cells, which are responsible for the production of pathogenic autoantibodies. This 'immune reset' approach is intended to halt the autoimmune process and potentially induce long-term remission in patients who have failed standard-of-care immunosuppressive therapies. It is currently being evaluated in Phase 1 clinical trials to assess safety, pharmacokinetics, and preliminary efficacy.
