This is a combination therapy that adds lumiliximab (an anti-CD23 chimeric monoclonal antibody) to the standard FCR regimen (fludarabine, cyclophosphamide, and rituximab) for the treatment of chronic lymphocytic leukemia (CLL). The combination was investigated based on preclinical data showing enhanced antitumor effects when lumiliximab was combined with fludarabine or rituximab. Initial phase 1/2 studies showed promising results with high complete response rates, but a subsequent randomized phase 2/3 trial (LUCID) failed to demonstrate significant improvement over FCR alone.
