Overview

LV20.19 CAR-T is a bispecific chimeric antigen receptor (CAR) T-cell therapy designed to simultaneously target CD19 and CD20 antigens on B-cell malignancies. Developed at the Medical College of Wisconsin, the therapy utilizes a lentiviral vector to engineer autologous T cells to express a single CAR construct with two distinct binding domains. This dual-targeting strategy is intended to mitigate the risk of disease relapse caused by antigen escape, which is a frequent limitation of monospecific CAR-T therapies targeting CD19 alone. LV20.19 CAR-T is currently being evaluated in Phase 1 and Phase 2 clinical trials for various B-cell lymphomas, including mantle cell lymphoma (MCL), chronic lymphocytic leukemia (CLL), and diffuse large B-cell lymphoma (DLBCL), both as a monotherapy and in combination with other agents such as the BTK inhibitor pirtobrutinib.
