LY4066434 is an oral, non-covalent, small molecule pan-KRAS inhibitor in early clinical development for the treatment of solid tumors harboring KRAS mutations. It is designed to target multiple KRAS mutant alleles—including G12C, G12D, G12V, G13D, G12A, and G12S—as well as wild-type KRAS while sparing HRAS and NRAS. By inhibiting mutant KRAS protein function and its downstream signaling pathways that drive cancer cell proliferation and survival (such as the MAPK/ERK pathway), LY4066434 aims to slow or stop tumor growth. The drug is being evaluated both as monotherapy and in combination with other anti-cancer agents in patients with advanced or metastatic solid tumors such as pancreatic cancer, colorectal cancer, non-small cell lung cancer (NSCLC), and others.
