m276-MMAE is an antibody-drug conjugate (ADC) targeting CD276 (also known as B7-H3), a cell surface glycoprotein that is highly overexpressed in a wide range of solid tumors and tumor-associated vasculature, while having limited expression in normal tissues. The ADC is composed of the human monoclonal antibody m276, which specifically binds to CD276, conjugated to the potent microtubule-disrupting agent monomethyl auristatin E (MMAE), typically via a protease-cleavable valine-citrulline (vc) linker. Upon binding to CD276 on the surface of target cells, the ADC is internalized via endocytosis and trafficked to lysosomes, where the MMAE payload is released. Once in the cytosol, MMAE binds to tubulin and inhibits its polymerization, leading to G2/M phase cell cycle arrest and apoptosis. Preclinical studies have demonstrated that m276-MMAE exerts potent antitumor activity by targeting both the malignant cells and the tumor microenvironment, including tumor-associated endothelial cells.
