Overview

m276-PBD is an antibody-drug conjugate (ADC) targeting CD276 (also known as B7-H3), a transmembrane protein frequently overexpressed on the surface of various solid tumors and their associated vasculature. The ADC consists of the m276 monoclonal antibody, which is humanized and uniquely cross-reactive with human, mouse, and rat CD276, conjugated to a pyrrolobenzodiazepine (PBD) dimer payload. The m276 antibody was specifically developed to facilitate preclinical evaluation in syngeneic mouse models. Upon binding to CD276 and subsequent internalization, the PBD dimer is released within the cell. The PBD dimer then binds to the minor groove of DNA and forms interstrand cross-links, leading to DNA damage and apoptosis. Developed by researchers at the National Cancer Institute, m276-PBD has demonstrated potent antitumor activity in preclinical models of several solid tumors, including those resistant to other ADC payloads.
