Overview

MB-CART20.1 is an autologous chimeric antigen receptor (CAR) T cell therapy that targets the CD20 antigen, a B-cell specific surface phosphoprotein expressed in B-cell lineage malignancies and certain melanoma subpopulations. The therapy consists of patient-derived (autologous) CD4+ and CD8+ T cells that are genetically modified ex vivo to express a second-generation anti-CD20 CAR containing the Leu16 scFv for specificity to CD20, fused with a 4-1BB costimulatory domain and a CD3ζ signaling domain[6][2][3]. Upon administration, these engineered T cells specifically recognize and kill tumor cells expressing the CD20 antigen through immunologic cytotoxicity[7][1]. MB-CART20.1 is being developed primarily for relapsed or refractory non-Hodgkin's lymphoma (NHL), but has also been investigated in malignant melanoma and precursor B-cell lymphoblastic leukemia/lymphoma[2][5].
