MDL-4102 is a first-in-class, highly selective small-molecule inhibitor of Bromodomain-containing protein 4 (BRD4) currently being developed by Model Medicines for undisclosed oncology indications. Discovered using the proprietary GALILEO AI-driven drug discovery platform through a high-throughput virtual screen, MDL-4102 is specifically engineered to overcome the dose-limiting toxicities typically observed with pan-BET (bromodomain and extra-terminal) inhibitors. It demonstrates high selectivity for BRD4 with no measurable activity against the related proteins BRD2 and BRD3. BRD4 is a critical epigenetic reader protein that facilitates the transcription of oncogenes and is implicated in both hematological and solid malignancies. The compound is currently in the preclinical stage (proof-of-concept), with plans to advance toward an Investigational New Drug (IND) submission in early 2027.
