MDX2004

MDX2004 is a tetraspecific T cell engager/expander multi-specific immune rejuvenator therapeutic being developed by ModeX Therapeutics, a subsidiary of OPKO Health. It is designed to target B-cell malignancies and autoimmune diseases by recognizing four targets: CD19, CD20, CD3, and CD28. This dual-tumor antigen and dual-T-cell pathway targeting is intended to overcome relapse and resistance in B-cell lymphomas and leukemias, with applications also under preclinical exploration for solid tumors, autoimmune disorders, and infections. The route of administration under evaluation is intravenous infusion. The precise mechanism of action is to promote T-cell activation, expansion, and survival, while simultaneously targeting malignant B cells through CD19 and CD20, aiming to maximize anti-tumor cytotoxicity and minimize tumor escape due to antigen loss. MDX2004 is currently in preclinical development, with clinical trials planned for late 2025 or early 2026[1][5][9].
