Overview

MEDI6469 + rituximab is a combination of two monoclonal antibodies. Rituximab is a chimeric monoclonal antibody that targets the CD20 antigen on B cells, leading to their depletion through mechanisms such as complement-dependent cytotoxicity and antibody-dependent cell-mediated cytotoxicity. It is used in the treatment of various B-cell malignancies (such as non-Hodgkin lymphoma and chronic lymphocytic leukemia) and autoimmune diseases (including rheumatoid arthritis, granulomatosis with polyangiitis, microscopic polyangiitis, and pemphigus vulgaris)[1][2][6][8]. MEDI6469 is an investigational anti-OX40 monoclonal antibody designed to stimulate T-cell activation by targeting the OX40 receptor (CD134), which plays a role in immune modulation. The combination aims to enhance anti-tumor immune responses by depleting B cells with rituximab while activating T cells with MEDI6469.
