Overview

This is a combination therapy of two immunosuppressive agents: - **Methylprednisolone** is a synthetic glucocorticoid corticosteroid with potent anti-inflammatory and immunosuppressive properties. It acts primarily by binding to the glucocorticoid receptor, modulating gene expression to suppress immune responses and inflammation. - **Mycophenolic acid** (often administered as its prodrug forms, mycophenolate mofetil or mycophenolate sodium) is an antimetabolite immunosuppressant that inhibits inosine monophosphate dehydrogenase (IMPDH), thereby blocking de novo guanosine nucleotide synthesis in lymphocytes. This results in selective inhibition of T and B lymphocyte proliferation. The combination is used off-label or as part of protocol-based regimens for various autoimmune diseases (such as systemic sclerosis, oral mucosal pemphigoid) and in transplant medicine to prevent organ rejection. The rationale for combining these drugs lies in their complementary mechanisms—methylprednisolone provides rapid suppression of inflammation while mycophenolic acid offers sustained inhibition of lymphocyte proliferation[1][4][5][6].
