MIDGE-CMVhTNF is a nonviral gene therapy vector developed by Mologen AG for the intratumoral expression of human tumor necrosis factor-alpha (hTNF-alpha). The vector utilizes the MIDGE (Minimalistic Immunologically Defined Gene Expression) platform, which consists of a small, double-stranded linear DNA molecule sealed with oligonucleotide loops, lacking the bacterial sequences and antibiotic resistance genes typically found in plasmid vectors. This specific construct incorporates a CMV promoter to drive the expression of hTNF-alpha. In preclinical studies, MIDGE-CMVhTNF has demonstrated significantly higher expression levels and improved safety profiles compared to traditional plasmid vectors, with rapid systemic clearance and localized activity following jet-injection into tumors such as melanoma.
