Overview

MIR143#12 is a chemically modified microRNA-143-3p (miR-143-3p) mimic developed to treat KRAS-mutant cancers. It is engineered for enhanced nuclease resistance and increased anti-cancer activity compared to unmodified miR-143. The drug acts by simultaneously silencing multiple nodes of the RAS signaling network, including KRAS, SOS1, AKT, and ERK. This broad suppression of the KRAS network induces apoptosis and inhibits cell growth more effectively than targeting KRAS alone, particularly in colorectal and pancreatic cancer models. Preclinical studies have also indicated that MIR143#12 may influence the tumor microenvironment by inducing neoangiogenesis while maintaining overall tumor growth suppression.
