Drug intelligence / Profile preview
MK-2690 (also known as SPR2015) is an investigational, preclinical, orally administered molecular glue inhibitor targeting the active, GTP-bound 'ON' state of the KRAS G12D mutant oncoprotein. Originally developed by SciBrunch Therapeutics, the compound was licensed globally by Merck & Co. in September 2026. In preclinical studies, MK-2690 demonstrated nanomolar antiproliferative activity across various KRAS G12D-mutant cancer cell lines while maintaining selectivity over wild-type KRAS. It also showed robust monotherapy antitumor efficacy in cell-derived and patient-derived xenograft (CDX/PDX) models. The drug is being developed for the treatment of KRAS G12D-driven cancers, including pancreatic, colorectal, and lung cancers.
