Overview
MK-8808 + cyclophosphamide + vincristine + prednisolone is a combination therapy under investigation for the treatment of diffuse large B-cell lymphoma. MK-8808 is a biosimilar of rituximab, which is a chimeric monoclonal antibody targeting CD20 on B lymphocytes, mediating B cell lysis through direct cytotoxicity and immune effector mechanisms such as antibody-dependent cell-mediated cytotoxicity and complement activation. Cyclophosphamide is an alkylating agent that interferes with DNA replication and transcription, leading to cell death, particularly in rapidly dividing cells. Vincristine is a vinca alkaloid that inhibits microtubule formation, arresting cells in metaphase and inducing apoptosis. Prednisolone is a synthetic glucocorticoid that modulates immune response and apoptosis through glucocorticoid receptor agonism. The combination is intended for use in B-cell malignancies, leveraging cytotoxic, immunomodulatory, and targeted B-cell depletion effects[3].
