Overview

mKRASvax is a synthetic long peptide vaccine designed to target multiple common mutations in the KRAS gene. It is formulated as a pooled set of peptides representing various mutant forms of KRAS and is administered with the poly-ICLC adjuvant to enhance immune response. The primary mechanism involves stimulating an immune response specifically against tumor cells harboring these KRAS mutations. The vaccine aims to induce T-cell mediated immunity capable of recognizing and attacking cancer cells expressing mutant KRAS proteins. It is being developed for use in combination with immune checkpoint inhibitors such as Balstilimab (anti-PD-1) and Botensilimab (anti-CTLA-4) for patients with unresectable or metastatic mismatch repair-proficient colorectal cancer (mCRC) and pancreatic ductal adenocarcinoma (PDAC), particularly after first-line chemotherapy[1][2][5][7]. Early clinical trials are evaluating its safety, immunogenicity, and preliminary efficacy.
