Monovalent anti-PD-1 refers to an antibody or antibody fragment (such as a Fab fragment) that binds to the Programmed Cell Death Protein 1 (PD-1) receptor with a single binding site. Unlike standard bivalent antibodies (e.g., nivolumab or pembrolizumab), monovalent versions block the interaction between PD-1 and its ligand PD-L1 but do not induce receptor crosslinking. Research indicates that this lack of crosslinking prevents PD-1 internalization and degradation, which is associated with reduced cytolytic activity and inferior anti-tumor responses in preclinical models. These molecules are primarily used as research tools to investigate the importance of PD-1 endocytosis in optimizing checkpoint blockade therapy.
