Overview

**Mosunetuzumab + tiragolumab** is an experimental combination therapy comprising two monoclonal antibodies—mosunetuzumab, a bispecific T-cell engager targeting CD20 on B cells and CD3 on T cells, and tiragolumab, an anti-TIGIT immune checkpoint inhibitor. The combination is under investigation for the treatment of relapsed or refractory B-cell non-Hodgkin lymphoma, specifically diffuse large B-cell lymphoma (DLBCL) and follicular lymphoma (FL). Mosunetuzumab redirects T cells to kill malignant B cells by bridging CD3 and CD20, promoting cytolytic synapse formation and T-cell mediated cytotoxicity. Tiragolumab targets TIGIT on T cells and NK cells, blocking its inhibitory signal and thereby enhancing antitumor immune responses. Both antibodies are developed by Hoffmann-La Roche/Roche, and the combination was evaluated in Phase 1/2 trials, now terminated, for these lymphomas[1][3][5][6][9].
