Overview

This combination therapy, evaluated in a Phase I/II trial at Northwestern University, integrates the redox-modulating agent motexafin gadolinium (Xcytrin) with the standard Zevalin radioimmunotherapy regimen for relapsed or refractory CD20-positive non-Hodgkin's lymphoma. The Zevalin regimen consists of the anti-CD20 murine monoclonal antibody ibritumomab tiuxetan, which is sequentially labeled with Indium-111 for biodistribution imaging and Yttrium-90 for targeted beta-radiation therapy, preceded by rituximab to optimize antibody distribution. Motexafin gadolinium acts as a radiosensitizer by disrupting cellular redox processes and increasing oxidative stress, thereby enhancing the cytotoxic effects of the Yttrium-90 radiation on lymphoma cells.
