Overview

MP0310 is a second-generation bispecific designed ankyrin repeat protein (DARPin) therapeutic developed for immuno-oncology applications. It is engineered to activate immune cells specifically within the tumor microenvironment by simultaneously binding to fibroblast activation protein (FAP), which is highly expressed on cancer-associated fibroblasts in solid tumors, and tumor necrosis factor receptor superfamily member 9 (4-1BB), an immune co-stimulatory receptor found on activated T lymphocytes and natural killer cells. This dual-targeting approach restricts T cell activation to tumor tissues, aiming to enhance anti-tumor efficacy while minimizing systemic toxicity. Preclinical studies have shown that MP0310 induces strong CD8 T cell activation and expansion only in the presence of both targets, leading to potent anti-tumor responses with reduced risk of systemic side effects compared to first-generation 4-1BB agonists. The drug was initially developed by Molecular Partners AG and has been evaluated in phase I clinical trials for advanced solid tumors[1][3][5][6][7].
