MRG001 is an investigational **CD20-directed antibody-drug conjugate** developed by Hangzhou Miracogen for relapsed or refractory B-cell non-Hodgkin lymphoma, including diffuse large B-cell lymphoma. It comprises a chimeric anti-CD20 monoclonal antibody linked through a protease-cleavable valine-citrulline linker to the microtubule-disrupting payload monomethyl auristatin E. Following CD20 binding and cellular internalization, intracellular linker cleavage releases the cytotoxic payload, resulting in tubulin disruption, mitotic arrest, and tumor-cell death. MRG001 has been evaluated in a first-in-human Phase 1 study in CD20-positive relapsed or refractory B-cell non-Hodgkin lymphoma and is being studied in Phase 2 combination treatment with orelabrutinib for relapsed or refractory diffuse large B-cell lymphoma.
